A five-year laboratory study found that fluoxetine—the active compound sold in the antidepressant Prozac—changed male guppies’ behaviour and reproductive traits and disrupted the relationships among those traits. The result is experimental evidence of harm in guppies, not proof that wild fish populations are undergoing reproductive collapse.
What happens when Prozac gets into waterways?
Wastewater can carry pharmaceutical residues into rivers and streams. In the study by Aich, Polverino and colleagues, guppies were exposed under controlled laboratory conditions to fluoxetine for five years across multiple generations. The researchers used progeny of wild-caught guppies (Poecilia reticulata) in 12 independent laboratory populations.
Fluoxetine is the chemical studied; Prozac is one brand name for it. The experiment tested mean measured concentrations of 0, 31.5 and 316 nanograms per litre (ng/L). The authors describe the treatment levels as field-relevant, but Monash University reported that water samples from the fish-collection site contained no fluoxetine. The experimental concentrations therefore must not be presented as measurements from that site.
What did the five-year guppy study measure?
The researchers repeatedly assessed male activity and risk-taking, then examined those behaviours alongside life-history and reproductive traits. Measurements included:

- sperm velocity.
This design allowed the team to study both average trait changes and whether traits that normally vary together remained coordinated.
What effects did fluoxetine exposure produce?
Disrupted links among traits
Long-term exposure altered associations among behavioural, life-history and reproductive traits. In other words, the exposure changed not only individual measurements but also the way characteristics covaried across fish. The paper discusses this pattern in relation to a “pace-of-life syndrome”—the coordinated suite of behavioural and life-history traits often used to describe how individuals allocate energy between activity, growth, survival and reproduction.

Less flexible behaviour
Exposed guppies showed reduced behavioural plasticity: they had less capacity to adjust activity and risk-taking when circumstances changed. Assistant Professor Giovanni Polverino of the University of Tuscia described the finding in Monash University’s 27 August 2024 account: “Fluoxetine exposure also significantly reduced the behavioural plasticity of guppies, leading to a lower capacity of the individuals to adjust their own activity and risk-taking behaviours across contexts,”
Changes in male reproductive traits
The university’s account highlights a combination of increased gonopodium size and reduced sperm velocity. Sperm velocity is one component of sperm performance; the study also measured sperm vitality and number. These observations should not be simplified to “the fish became infertile.” They indicate changes in reproductive traits under the tested exposure regime.

The concentrations in this table describe the study treatments. They are not estimates of the number of wild fish affected and were not reported as measurements from the collection-site water.
Does Prozac pollution affect fish reproduction in the wild?
The study shows that sustained fluoxetine exposure can affect guppy behaviour and male reproductive characteristics in a laboratory setting. It does not establish that wild guppy populations—or fish populations generally—are experiencing reproductive collapse, declining abundance or failure to replace themselves.

Wild populations face changing concentrations, seasonal conditions, mixtures of pollutants, predators, food availability and movement between habitats. None of those variables can be inferred from this experiment alone. The sources available for this study also do not provide a named population-level statistic for reproductive decline in wild fish or a regulator-established safe environmental threshold for fluoxetine.

Reproduction depends on interacting traits rather than a single measurement. A fish may alter its activity, risk-taking, body condition, sexual signal or sperm performance, while the usual links among those traits also change. Reduced plasticity could matter because flexible behaviour helps animals respond to different contexts, but the guppy experiment did not measure long-term population growth in a natural waterway.

The strongest conclusion is therefore specific: multigenerational fluoxetine exposure changed the behavioural and reproductive phenotype of male guppies and disrupted coordination among traits. Whether comparable effects occur in particular wild populations requires separate field measurements and population-level studies.
