Did Scientists Really Revive the Dire Wolf? What Colossal Made

Colossal’s three “dire wolves” are cloned gray wolves edited at 20 genetic sites to carry selected dire-wolf traits. Here’s what the science does—and does not—show.

Not literally. Colossal Biosciences produced three cloned gray wolves with 20 targeted genetic edits intended to give them selected dire-wolf traits. They are living animals, but they are not genetic copies of the extinct dire wolf. The IUCN SSC Canid Specialist Group says they are not dire wolves; Colossal calls its work de-extinction.

What did Colossal actually create?

Colossal announced three animals—Romulus, Remus and Khaleesi—as dire wolves. The more precise description used by independent and institutional sources is genetically modified gray wolves carrying selected dire-wolf traits. Romulus and Remus were born on October 1, 2024, and Khaleesi on January 30, 2025, according to Colossal.

The distinction is not just a matter of naming. The extinct dire wolf and the living gray wolf are different species, and changing selected parts of one species’ genome does not turn it into an exact copy of another. UC Santa Cruz says the resulting animals are not exact replicas of their ancestors. The IUCN SSC Canid Specialist Group states that “the three animals produced by Colossal are not dire wolves.”

How were the edited wolf pups made?

Colossal reports 20 targeted edits across 14 genes. The work used ancient dire-wolf DNA to identify candidate sequences, but did not splice ancient dire-wolf DNA into the gray-wolf genome. Instead, researchers rewrote selected gray-wolf genes to match chosen dire-wolf sequences.

  1. Study ancient remains: Colossal says its genomic references came from a 13,000-year-old dire-wolf tooth from Ohio and a 72,000-year-old ear bone from Idaho.
  2. Edit gray-wolf cells: Researchers used endothelial progenitor cells taken from gray-wolf blood and made the targeted edits in those cells.
  3. Clone embryos: They transferred nuclei from edited cells into eggs whose own nuclei had been removed.
  4. Implant embryos: The embryos were implanted into domestic-dog surrogate mothers. The Associated Press reported pregnancies lasting 62 days before the pups were born.

That is cloning combined with genetic editing—not a process that reconstructs an extinct animal’s full genome. The edits were made at selected sites, not across all the genetic differences between the two species.

Why is “de-extinction” disputed?

“De-extinction” can describe an effort to recover some characteristics of an extinct species, but it can also sound like the original species has been restored. In this case, the company’s label and the biological description do not mean the same thing: Colossal presents the animals as dire wolves, while the IUCN SSC Canid Specialist Group and independent scientists describe them as edited gray wolves.

The IUCN group says dire wolves and gray wolves differ at thousands of genes, whereas Colossal edited a handful of genes. Colossal’s chief science officer Beth Shapiro has also acknowledged the distinction: “It’s not possible to bring something back that is identical to a species that used to be alive. Our animals are grey wolves with 20 edits that are cloned.” Paleogeneticist Nic Lawrence of the University of Otago similarly said that a gray wolf with 20 edits to 14 genes “is still very much a gray wolf.”

University at Buffalo biologist Vincent Lynch, quoted by the Associated Press, put the broader limitation this way: “All you can do now is make something look superficially like something else”—not fully revive an extinct species. The animals may display selected traits associated with dire wolves, but resemblance and selected genetic changes do not establish that the extinct species itself has returned.

What did ancient DNA reveal about dire wolves?

The ancient samples were used as genomic references, and the resulting research also sheds light on dire-wolf ancestry. UC Santa Cruz reports that roughly two-thirds of the dire-wolf genome appears to descend from an ancestor related to wolves and coyotes, while the remaining third comes from an ancient lineage related to South American dogs. Researchers infer that dire wolves arose through hybrid speciation around 4–5 million years ago.

That finding concerns the evolutionary history of the extinct species; it does not mean the new animals were made by combining living wolves, coyotes and South American dogs. Colossal’s described method was to edit gray-wolf cells at selected sites using sequences identified from ancient dire-wolf genomes.

Could these animals live in the wild or restore dire-wolf ecology?

The available evidence does not establish that the edited wolves can or should live in the wild. Nor does producing animals with selected dire-wolf traits demonstrate that an extinct ecosystem has been restored.

The dire wolf disappeared about 12,000 years ago. The IUCN SSC Canid Specialist Group notes that the ecological setting and prey of that period—including mammoths and giant ground sloths—are gone, while gray wolves already occupy a modern ecological niche. A genetic proxy cannot bring back those vanished ecological relationships. The group warns that creating proxy animals does not restore ecosystem function and could draw attention or resources away from habitat protection and the recovery of living species.

Those limits matter especially when discussing animals produced through cloning and genetic editing. A claim that a proxy could be released, survive in a particular habitat, or provide conservation benefits would require evidence about that specific animal and setting. The material available here does not establish such an outcome.

How does this differ from cloning for conservation?

The same general cloning platform may have a separate conservation application. The Associated Press reports that Colossal cloned four red wolves using blood from wild wolves, with the aim of adding genetic diversity to captive breeding and recovery efforts. That work concerns an endangered living species and is distinct from the dire-wolf demonstration: it does not show that an extinct species has been restored.

For conservation, the relevant question is whether a technique helps a living population recover—not whether an animal resembles an extinct one. The reported red-wolf effort has that stated aim, but the information here does not establish its eventual effect on the population.
