Provora: the 2022 eukaryotic supergroup unlike other known organisms

Provora is a 2022 eukaryotic supergroup of rare microbial predators. Researchers described ten cultured strains in two clades, Nebulidia and Nibbleridia, distinguished by genes, cell structure and behavior.

Provora is a supergroup of microscopic, predatory eukaryotes described in a Nature paper published on 7 December 2022. The study reported ten previously undescribed strains grown in culture and placed them in two deeply divergent clades, Nebulidia and Nibbleridia. The authors found that Provora differs from other eukaryotes in genes, cell structure and behavior.

What is Provora?

Provora is a proposed supergroup within the eukaryotes—the broad group whose cells contain nuclei. It is not a newly discovered domain separate from all known life, and the study did not claim that these organisms are extraterrestrial or non-eukaryotic. “New branch on the Tree of Life” is a headline shorthand for a major evolutionary grouping recognized from multiple lines of evidence.

The organisms are microscopic predators. They were identified through culture-based isolation rather than only from DNA sequences collected directly from the environment.

What the 2022 study actually found

Ten cultured strains

Tikhonenkov and colleagues described ten previously undescribed predator strains. That is the number of strains reported in this study—not a count of all Provora species, individuals or populations on Earth. The paper’s abstract does not provide a global population estimate.

Two contrasting clades

The two groups can look superficially similar, but the authors report fundamental differences in ultrastructure, behavior and gene content. Those differences, together with genetic analyses, support treating them as separate deep lineages within Provora.

Why scientists consider Provora unusually distinct

Genetic evidence

Comparative genetic analyses placed the strains in a lineage distinct from other sampled eukaryotes. Their gene content also differs between Nebulidia and Nibbleridia, adding evidence that the apparent similarity between the clades does not reflect close overall identity.

Cell structure

Microscopic and ultrastructural observations revealed distinctive cellular organization. “Unlike any others on Earth” is therefore best read as a summary of the paper’s finding that Provora is genetically, morphologically and behaviorally distinct from other eukaryotes—not as a claim that every feature is entirely unprecedented.

Predatory behavior

Provora feed as microbial predators. Their behavior supplied another independent feature separating the group from known eukaryotic lineages.

Where Provora lives

The study reports Provora from both marine and freshwater environments and describes the organisms as numerically rare. Because they occur at low abundance, routine environmental DNA surveys may miss them or produce too little information to reveal their biology. The paper’s reported habitats are broad; they do not establish a single location, range size or worldwide population total.

Why culture was important

Researchers first had to isolate the organisms and maintain them in culture to study their cells, feeding behavior and genetics together. High-throughput sequencing can detect enormous numbers of environmental DNA fragments, but a sequence alone may not show what an organism looks like, how it behaves or how it obtains food.

The study authors summarized the point in the Nature abstract: “In the age of high-throughput analyses, investigation of eukaryotic diversity through culture remains indispensable for the discovery of rare but ecologically and evolutionarily important eukaryotes.” Culture and environmental sequencing are complementary approaches: one provides detailed biological observations, while the other can reveal diversity that is difficult to grow.

What “a new branch” does—and does not—mean

  • It means: the authors proposed a previously unrecognized supergroup supported by genetic, structural and behavioral evidence.
  • It does not mean: scientists found a new domain of life, counted every Provora organism, or showed that the organisms are unrelated to all other eukaryotes.
  • The ten-strain figure means: ten strains were described in the 2022 study; it is not a census of Provora diversity in nature.

Why the discovery matters

Provora demonstrates how much eukaryotic diversity can remain hidden when organisms are rare and difficult to grow. It also shows why evolutionary classification cannot rely on appearance alone: the two Provora clades may seem similar at first glance while differing in internal structure, genes and behavior. More sampling and additional isolates would be needed to determine how much diversity exists within the supergroup and how widespread its members are.
