It can cause marked sedation, but quietness does not mean a dog’s pain has been treated or that fear has been resolved. Give it only as directed by the prescribing veterinarian; an online dose is not a safe basis for medicating your dog.
What acepromazine does—and what it does not do
Acepromazine maleate depresses the central nervous system. The U.S. PROMACE tablet label describes its effects as sedation, muscle relaxation, and reduced spontaneous activity, and its veterinary uses as an aid in tranquilization and preanesthetic use in dogs, including some examinations and procedures. It is not a pain reliever: UK product information describes its analgesic effects as negligible. A dog that is still or easier to handle may still be experiencing pain and may need separate pain control.
Sedation also does not establish that fear has been adequately treated. Response varies, and the UK product information notes that some animals may not achieve reliable sedation. Ask the veterinarian what outcome is intended and what monitoring or additional treatment is appropriate for your dog.
Why you should not use an online dose
The U.S. PROMACE tablet label lists a dosage range of 0.25–1.0 mg per pound and says it may be repeated as required. That is label information, not an individualized recommendation: the same label states, “Federal law restricts this drug to use by or on the order of a licensed veterinarian.” Do not calculate a dose, change the amount, or repeat it based on a website or a previous prescription. The right instructions depend on the specific prescription and your dog’s circumstances.

Before giving a prescribed dose, confirm the amount, timing, whether any repeat dose is authorized, and what to do if the expected effect does not occur. Tell the veterinarian about all other medicines and your dog’s medical history. Risks, cautions, and situations to discuss with the veterinarian
Sedation can be marked or prolonged
The U.S. label warns that sensitive animals or excessive amounts may experience prolonged depression or motor restlessness. Acepromazine’s effects can add to those of other central nervous system depressants and can potentiate general anesthesia. Make sure the veterinarian knows about every medication or sedative your dog has received, including products used for parasites.

Health conditions and interacting products matter
The U.S. tablet label calls for greater care in dogs experiencing stress, debilitation, cardiac disease, sympathetic blockade, hypovolemia, or shock. It also advises caution with a history of liver dysfunction or leukopenia. It contraindicates use with organophosphorus wormers or ectoparasiticides—including flea collars—and with procaine hydrochloride. The label says not to use acepromazine to control tremors from organophosphate poisoning.
Temperature, blood pressure, and breed or genetic factors
The UK Veterinary Medicines Directorate’s product information, revised August 2025, identifies hypotension and possible hypothermia. It reports that dogs with the ABCB1-1Δ (MDR1) mutation may have more profound and prolonged sedation, and that large breeds may be particularly sensitive. That UK product summary also recommends dose reduction for dogs with the mutation; it is not a U.S. dosing instruction. Tell your veterinarian about any known mutation or breed-related concern and ask how your dog will be monitored.

What to expect and how to respond
The evidence cited here does not establish a dependable duration for acepromazine’s effects in every dog, so a universal answer to “how long does it last?” would be misleading. The U.S. label warns that sedation can be prolonged, but your veterinarian is the right person to explain the expected course for the prescribed product, dose, and dog.
- Before administration: Confirm the prescribed dose and timing, any repeat-dose instruction, relevant health conditions, and possible interactions with the veterinary team.
- After administration: Follow the veterinarian’s instructions for observation and contact them if the response concerns you or differs from what they described.
- If a reaction or accidental ingestion is suspected: Contact your veterinarian or an emergency veterinary service promptly. Do not try an at-home treatment based on guesswork.
The U.S. label describes a manufacturer safety study in which dogs received three times the upper limit of its recommended daily dosage (3.0 mg/lb); it reports mild depression that disappeared in most dogs within 24 hours after dosing stopped. This is a label-study description, not a general risk estimate or reassurance that an excessive dose is safe for an individual dog.

Discussing sedation options for an exam or procedure
If the concern is that sedation alone may not address pain, ask the veterinarian whether the procedure requires separate analgesia or whether another approach is suitable. One example—not a recommendation for every dog—is Zenalpha (medetomidine and vatinoxan), which FDA reported approving on March 30, 2022, as a veterinarian-administered sedative and analgesic for dogs undergoing examinations or certain procedures. FDA says it is for licensed-veterinarian use with vital-sign monitoring. In the Zenalpha study or label context, FDA reported diarrhea, muscle tremors, and colitis in 2 to 3 percent of treated dogs; these are not acepromazine statistics. The FDA comparison was with dexmedetomidine, not acepromazine, so it does not establish that Zenalpha is superior to acepromazine. The veterinarian must assess whether any option fits your dog.
