To collect a usable dog DNA sample at home, follow the exact instructions for your kit or laboratory: accepted swabs, food-and-water waiting time, handling, drying, and packaging can all differ. Keep your dog away from other pets and shared mouth-contact items for the period specified, wash your hands, and touch only the swab or brush handle. A clean collection depends on using the provider’s method—not on improvising with a different swab or packaging.
Before you open the kit: check its exact protocol
Read the complete instructions first and confirm what collection device the provider accepts. Consumer kits and veterinary laboratories may use different materials and workflows. Embark instructs customers to use the swab included in its kit; UC Davis Veterinary Genetics Laboratory (VGL) specifies three cytology brushes or four accepted interdental brushes for its dog samples. Do not substitute one type for another unless your provider explicitly permits it.

How long should your dog wait after eating or drinking?
Use the waiting period given by your own kit or laboratory. Official instructions differ, so there is no single interval that applies to every test.

These are provider-specific procedures, not interchangeable recommendations. PennGen also says to inspect for visible food debris; if debris is present, its instructions advise cleaning or rinsing the mouth and waiting another hour. Apply that step only if it matches the protocol for your test.

Collect the sample without introducing other DNA
- Prepare one dog at a time. Keep the dog away from other pets and shared toys for the time specified by the provider. If you are sampling several dogs, finish and package one dog’s sample before starting the next.
- Clean your hands. Wash them or use clean disposable gloves, as the provider directs. Set the unopened collection materials on a clean surface.
- Open the package as instructed. Hold the swab or brush by its handle. Do not touch the collection tip or bristles, and keep them away from people, other animals, and surfaces.
- Use the provider’s collection technique. For the UC Davis brush method, place the bristle head between the gum and cheek, press lightly from outside the cheek, and rotate or rub for 15 seconds. Its target is cheek cells, not drool alone; it says not to rub the brush on the tongue or teeth.
- Use the specified number of collection tools. Follow the kit or lab’s count and method. When moving between dogs, wash your hands or change gloves.
- Check the identity and complete the paperwork. Match each sample to the correct dog before packaging and confirm the required identifying details on the label and submission form. This is especially important for similar-looking littermates.
UC Davis emphasizes: “Do not collect saliva/drool – the key to obtaining a good sample is getting cheek cells on the swab.” Follow a different technique if your particular kit gives different instructions.
Drying, packaging, and shipping are not universal
Keep each sample in the collection system supplied or approved by its provider. For example, UC Davis instructs owners to air-dry brushes and leave them unsealed in their sleeve; PennGen calls for complete air-drying and returning the brush to its package; Embark’s consumer kit uses its supplied collection tube. These directions describe different protocols, not steps to combine.

- Do not put a consumer-kit swab into a laboratory brush envelope, or place a lab brush in a consumer tube.
- Follow the provider’s directions for drying, sealing, labeling, forms, and shipping.
- If the instructions are unclear or a swab touches a surface, contact the provider before mailing the sample rather than guessing whether it remains usable.

Extra precautions for puppies and multi-dog households
Puppies that still nurse
A puppy’s mouth may contain the mother’s DNA after nursing. UC Davis recommends waiting until a puppy is at least three weeks old; PennGen says not to use cheek brushes on unweaned puppies. Because the advice and acceptable procedure depend on the test, ask the provider or your veterinarian before collecting from a young puppy.

Several dogs or shared equipment
Collect and package each dog’s sample separately, check its label immediately, and clean your hands or change gloves before handling the next dog’s materials. PennGen warns that even minuscule contamination from another animal or person could lead to false results; its instructions also caution about mislabeling, particularly when littermates look alike.

A cheek swab may yield less DNA or lower-quality DNA than a blood sample, according to Colorado State University’s Webb Laboratory. An inadequate sample may require recollection, but the cited providers do not give a general failure rate. Ask your test provider whether it wants a new cheek sample or a different sample type rather than switching methods on your own.
