Do Icefish Have High Cholesterol in Their Cell Membranes?

Icefish do not all have high membrane cholesterol. Findings vary by species and tissue, and cholesterol is only one factor affecting membrane fluidity.

No—not as a group. Cholesterol levels differ by icefish species, tissue and membrane type. In one Antarctic heart-membrane comparison, the icefish Champsocephalus gunnari had the lowest cholesterol measured, while two red-blooded Antarctic fishes had the highest. Other measurements show that some icefish membranes can have cholesterol similar to, or higher than, a particular red-blooded comparator.

Why there is no single answer for all icefish

“Icefish” covers multiple species, and a cell does not have just one membrane composition throughout its body. Studies have measured different organs and membrane fractions—including heart membranes, synaptic membranes, myelin, gill plasma membranes and cardiac microsomes. Their findings cannot be collapsed into one cholesterol level for every icefish cell.

The comparison fish matter, too. Several studies used the red-blooded Antarctic notothenioid Notothenia coriiceps; another heart study compared four Antarctic notothenioids. A result that is higher than N. coriiceps in one tissue does not establish that all icefish membranes have high cholesterol.

What comparisons of icefish membranes found

Heart membranes from four Antarctic notothenioids C. gunnari, an icefish, had the lowest cholesterol; the highest values were in the red-blooded Gobionotothen gibberifrons and N. coriiceps. All four icefish had more fluid membranes than N. coriiceps. Icefish had about 1.5 times as much phosphatidylethanolamine (PE) as the red-blooded fishes in this comparison.
Neuronal membranes of the icefish Chaenocephalus aceratus and N. coriiceps Synaptic cholesterol was similar in the two species. Myelin cholesterol was lower in N. coriiceps than in the icefish. C. aceratus synaptic membranes were consistently more fluid. The study authors suggested that similar synaptic cholesterol could help explain comparable Na+/K+-ATPase activity.
Gill plasma membranes, cardiac microsomes, synaptic membranes and myelin in N. coriiceps acclimated from 0°C to 5°C Cholesterol rose 1.2-fold in gill plasma membranes and 1.6-fold in cardiac microsomes. It did not change in synaptic membranes or myelin. The response varied by tissue and membrane fraction; this result concerns a red-blooded species, not a general icefish trait.
Liver membranes in an Antarctic species comparison that included the icefish Chionodraco hamatus Cholesterol varied more with species than with habitat temperature. It was higher in the New Zealand fish than in some Antarctic species, but not different from every Antarctic icefish. The comparison supports species-specific variation rather than a simple temperature-based rule.

How icefish membranes stay functional in cold water

Cholesterol is only one part of membrane composition. Phospholipid classes and the fatty acids in those lipids also influence membrane fluidity. The heart comparison, for example, found both greater fluidity and more PE in icefish than in N. coriiceps, while the neuronal comparison found that fluidity and cholesterol did not move in lockstep across membrane types.

That is why “more cholesterol prevents membranes from becoming too fluid” is not an adequate general explanation for icefish. The measurements show different combinations across species and tissues, rather than a single cholesterol adjustment shared by all icefish.

What temperature acclimation tells us—and what it does not

In N. coriiceps, moving from 0°C to 5°C was associated with cholesterol increases in gill plasma membranes and cardiac microsomes, but not in synaptic membranes or myelin. This tissue-specific response shows that temperature acclimation can affect membrane cholesterol in some contexts. It does not show that all icefish have high cholesterol, nor does it establish a universal cold-adaptation mechanism.

A 2018 comparative study by Vanita C. Malekar and coauthors described its findings as “the first study to show a homeoviscous response to higher temperatures in an Antarctic fish, although for only one of the two species examined.” The qualification matters: the result was limited to the species and conditions studied.

How to interpret claims about “high” membrane cholesterol

  • Check the species: an icefish result from C. gunnari, C. aceratus or C. hamatus should not automatically be applied to other icefish.
  • Check the tissue and membrane fraction: heart, gill, synapse and myelin measurements are not interchangeable.
  • Check the comparator: “higher” or “lower” only has meaning relative to the species and tissue measured in that study.
  • Consider the rest of the membrane: phospholipid composition and fatty-acid unsaturation also matter for fluidity.

Published studies cover selected species, tissues and experimental designs, not every icefish cell membrane. No single cholesterol value represents the whole group.
