The dire wolf has not been brought back from extinction in the strict biological sense. Colossal Biosciences produced three wolf pups—Romulus, Remus and Khaleesi—by editing cells from living gray wolves and cloning those edited cells. Ancient dire-wolf DNA informed the research, but no complete ancient genome or dire-wolf cell was placed into an embryo.
The pups are best described as genetically modified gray wolves designed to resemble dire wolves in selected traits. That is a major biotechnology achievement, but it is not the same as cloning an extinct species from ancient DNA.
What scientists actually created
Colossal announced the animals in April 2025. Romulus and Remus were born on October 1, 2024, and Khaleesi was born on January 30, 2025. The company calls them the first “de-extinct” dire wolves, while several researchers—including Colossal’s chief science officer and ancient-DNA researcher Beth Shapiro—have described them more precisely as cloned gray wolves carrying 20 genetic edits.
The process began with endothelial progenitor cells taken from living gray wolves. Researchers edited those cells at 20 targeted locations spanning 14 genes. They then used nuclei from the edited cells to create embryos through cloning technology. Domestic dogs carried the embryos as surrogate mothers.

That means the animals’ underlying genome is still that of a gray wolf. The edits were intended to produce features associated with the extinct dire wolf, including:
- a larger head and teeth;
- broader shoulders and more muscular legs;
- a white coat;
- large adult-size projections; and
- some changes related to vocalization.
The animals may look substantially different from ordinary gray wolves, but appearance alone cannot establish that they belong to the extinct species Aenocyon dirus.
Why ancient DNA mattered—but was not used as a blueprint
Ancient DNA recovered from fossils is fragmented and incomplete; scientists cannot simply extract an intact dire-wolf genome and insert it into an egg. No complete living dire-wolf cell was available to clone, either.
Researchers analyzed DNA from dire-wolf remains, including fossils more than 13,000 and 72,000 years old. This paleogenomic work helped illuminate the animal’s evolutionary history and identify genetic differences that could inform selected edits associated with traits such as size, skull shape, muscle development, coat color and vocalization. It did not make the pups’ genome a dire-wolf genome.

The ancient genome data therefore served as a research reference. It did not become the genome of any of the three pups.
Why these are not simply ancient gray wolves
The extinct dire wolf was not a recent variety of modern gray wolf. A peer-reviewed study published in Nature found that dire wolves separated from the lineage leading to living gray wolves and related canids roughly 5.7 million years ago, with no evidence of recent gene flow between dire wolves and modern gray wolves or coyotes.
That evolutionary distance matters. A dire wolf and a gray wolf may share many genes because both are canids, but a species’ biology is not defined by a short list of visible features. It also depends on the full genome, including regulatory DNA that controls when and where genes work.
Changing 20 sites cannot recreate every dire-wolf characteristic. The edits do not reproduce:

- its embryonic and fetal development;
- the maternal environment in which it evolved;
- its learned behavior and social patterns;
- its microbiome and immune history; or
- its relationships with the Ice Age ecosystem and prey.
The often-repeated claim that gray wolves and dire wolves are about “99.5% identical” needs context. Even a small percentage difference across a large mammalian genome can represent millions of DNA differences. Some occur in regulatory regions whose effects may not be obvious from appearance.
Clone, genetically modified animal and de-extinct species: the distinction

Why the wording is controversial
“Dire wolf” is being used in two different ways. Colossal uses it as a practical or colloquial label for animals engineered to express selected dire-wolf traits. In taxonomy, however, the name refers to an extinct evolutionary lineage, Aenocyon dirus.
Those meanings are not interchangeable. A white, large-headed wolf with enlarged teeth can resemble a reconstruction of a dire wolf without being a resurrected member of that species. The most scientifically cautious description is: three cloned, genetically modified gray wolves engineered to express selected traits inferred from ancient dire-wolf DNA.
This wording does not diminish the technical accomplishment. Editing multiple genes, producing viable cloned embryos and raising healthy animals are difficult tasks. It separates what the experiment demonstrated from the larger claim that an extinct species has returned.

Are the pups pets, or could people own one?
No. These animals are not pets and are not being offered for private ownership. Colossal says Romulus, Remus and Khaleesi live at a secure 2,000-acre ecological preserve in the United States under continuous veterinary monitoring. The company says they are not capable of living in the wild and are not being prepared for release.
Regardless of their appearance, they remain large, powerful wild canids with specialized care requirements. A dire-wolf-like animal would not be equivalent to a large dog breed, and selective breeding for a dramatic appearance does not create domestication.
For pet owners, the practical lesson is straightforward: a wolf, wolf hybrid or engineered wolf-like animal is not a realistic household companion. Such an animal would raise serious safety, welfare and legal concerns, as well as housing and veterinary needs far beyond those of a dog.

The project could contribute methods relevant to conservation biology. Genome editing and cloning may eventually help researchers preserve genetic diversity in endangered species, study disease resistance or support reproduction when few breeding animals remain. The ancient-DNA research has also improved scientists’ understanding of canid evolution.

But producing a few animals with traits associated with an extinct species does not restore an ecosystem. Dire wolves disappeared thousands of years ago, along with the ecological conditions in which they lived. A laboratory-created proxy would not automatically fill the same role, interact with prey in the same way or repair the causes of extinction.
Animal welfare, ecological risk, genetic safety and control of the technology are separate questions from whether the pups look impressive. They need careful evaluation if similar methods are used in conservation projects.
What the headline gets wrong
- They were not cloned from an extinct dire wolf. The cloned material came from edited cells of living gray wolves.
- Ancient DNA informed the research but did not become the pups’ complete genome. Fossil DNA was analyzed to study dire-wolf genetics and guide selected edits.
- The pups are not universally accepted as members of Aenocyon dirus. Many scientists would call them genetically modified gray wolves or engineered proxies.
- The animals are not pets or candidates for release. Colossal says they remain under managed care at a secure preserve and are not being prepared for release.
