What did the monkey study find?
Castner and colleagues reported in Nature Aging on July 3, 2023, that a single subcutaneous dose of klotho improved aged rhesus macaques’ performance on a spatial delayed-response task. The authors summarized the result this way: “a single administration of low-dose, but not high-dose, klotho enhanced memory in aged nonhuman primates.” Read the study in Nature Aging.

The study included 18 macaques, 13 females and five males, aged 15 to 28 years; their average age was about 21.78 years. Researchers generated the rhesus macaque form of klotho and administered it by injection under the skin.

In a spatial delayed-response task, each monkey had to remember where a food reward had been hidden. Researchers tested versions with normal and high memory loads, which differed in how demanding the memory task was. The measured outcome was performance on these tasks—not a broad measure of memory, everyday function, or biological aging.

How much klotho did the monkeys receive, and how long did the effect last?
The main cognitive analysis used a single 10 μg/kg dose. Improvement was reported beginning four hours after treatment on the high-memory-load task, with improvement across both task versions. The reported effect persisted over the two-week testing period, meaning at least two weeks in this experiment.
The researchers also explored 20 and 30 μg/kg doses; neither showed significant cognitive improvement in the reported tasks. These experimental doses are not treatment recommendations for monkeys or people.

Does this mean there is an anti-aging injection for people?
No. This was preclinical research in nonhuman primates. It does not show that klotho improves memory in people, establish long-term safety or a clinically useful human dose, or demonstrate that a klotho treatment is available. The authors said systemic low-dose klotho “may prove therapeutic in aging humans”; that is a possibility they proposed, not a finding from human treatment.

The study’s disclosures also provide relevant context: the Regents of the University of California held an issued patent on methods and compositions involving klotho and improved cognition, and the Regents, Yale University, and Unity Biotechnology had applied for a provisional patent involving klotho and cognition. Some authors disclosed consulting, stock holdings, or company-founding interests. These disclosures do not by themselves settle the validity of the findings, but they are part of the context in which to assess them.
