Some studies report improved embryo development or semen-quality measures under specific protocols, but that is not proof of better pregnancy rates, herd health, or productivity overall. Other results are mixed, and the evidence does not establish a general cattle treatment or a ready-to-use dose.
Why scientists are studying melatonin in cattle
Melatonin is a hormone associated with biological rhythms. Bovine reproductive research has examined its possible roles in ovarian cells, oocyte development, embryo production, testicular function, sperm quality, stress, and inflammation. Researchers are also interested in its antioxidant properties and how seasonal conditions, including day length and heat, may interact with reproductive biology.
A 2023 review of bovine reproductive biotechnology describes the field as promising while noting significant challenges to safe and effective application. A broader 2026 review discusses possible roles in immunity, reproduction, metabolism, and environmental stress responses across livestock species. That wider review offers context, not cattle-specific evidence for a product, dose, or treatment protocol.
These subjects span very different levels of evidence. A change in cells or gene expression is not the same as more embryos, a higher pregnancy rate, improved milk production, or healthier calves. The practical importance depends on what a study measured, how it administered melatonin, and which animals and conditions it tested.

What cattle studies have found
| Study and setting | Reported result | What the result does—and does not—show |
|---|---|---|
| Solanki and colleagues, 2026: 25 non-lactating Sahiwal cows received an intramuscular melatonin protocol and 19 received placebo before ovum pickup in summer. | In-vitro blastocyst development was 38.41% in the treated group and 25.29% in controls (P < 0.04). | This is an embryo-development result under a defined summer protocol. It does not establish improved pregnancy rates or routine herd productivity. |
| Isobe, Yoshimura, and Ikebata, 2026: Japanese Black cows studied during the hot season in ovum-pickup/in-vitro production and embryo-recovery experiments. | The reported rate ratio for freezable embryos was 2.85 in one experiment after two injections and 1.48 in a second experiment after injections on two days. One injection alone had no effect in the second experiment. | The effects varied with the protocol and experiment. Pregnancy outcomes among repeat-breeder cows only tended to improve, and the researchers said practical effects under heat stress remain unclear. |
| 2023 spring-calving beef-cattle study: prenatal and postnatal implantation. | Prenatal treatment did not affect calf birth weight. Neither prenatal nor postnatal treatment changed weaning weight or later bull scrotal/testicular measures. Dam milk yield and fat percentage tended to decrease in the prenatal melatonin group. | The study did not find improved offspring performance on the reported measures. USDA’s interpretive summary noted that seasonal differences in day length and temperature may have affected the dams’ response. |
| 2024 study of six Murrah buffalo bulls, three per group, comparing implanted animals with controls. | The implanted group had better post-thaw motility and viability measures in the post-implantation period. First-service pregnancy rates were 43.3% versus 30.0%, a difference that was not statistically significant. | This small study involved buffalo, not cattle. It suggests a semen-quality finding in that group, but does not establish a fertility benefit for cattle herds. |
| 2013 study of high-producing dairy cows treated during early dry-off under heat-stress conditions. | The paper reported improved subsequent reproductive performance. | This is a study-specific result; it does not resolve effects across breeds, seasons, or management settings. |
Taken together, the studies point to a possible research opportunity in reproductive biology, not a consistent production effect. Even a statistically significant improvement in a laboratory endpoint does not by itself demonstrate that cows will conceive more often, that more calves will be born, or that a treatment will be worthwhile under commercial herd conditions.
How to interpret the different outcomes
- Cell and mechanism findings: These can help explain how melatonin might act, but they do not show that a whole animal or herd benefits.
- Embryo development and freezable embryo counts: These are relevant to reproductive biotechnology and embryo production. They are not equivalent to confirmed pregnancies or more calves born.
- Semen measures: Post-thaw motility and viability can indicate sample quality, but they do not guarantee successful breeding. The buffalo study’s first-service pregnancy difference was not statistically significant.
- Animal and production outcomes: Pregnancy, milk yield, calf growth, and later fertility are closer to farm-level outcomes. The reported beef-cattle study did not find improvement in the offspring measures it tracked and noted a tendency toward lower milk yield and fat in treated dams.
When comparing a melatonin study with another, check the animal type, production purpose, season and heat exposure, route and timing, sample size, and endpoint. A result in non-lactating Sahiwal donor cows during summer cannot automatically be applied to a lactating dairy herd, a spring-calving beef operation, or another breed.

Does cattle research establish a treatment or dose?
No reader-ready dose or treatment recommendation is established by these studies. Published protocols include injections and subdermal implants, with timing and amounts that differ by experiment. Those are research protocols, not consumer directions, and the results cannot be used to infer that a human sleep supplement, research formulation, or generic implant tool is an equivalent cattle treatment.
Product status is a separate question from whether an experimental result is promising. The U.S. Food and Drug Administration states: “To legally market an animal drug, a drug company must get the drug approved, conditionally approved, or indexed by FDA.” FDA explains that most approved animal drugs can be checked through Animal Drugs @ FDA and the Green Book; a National Drug Code by itself does not establish that a drug is legally marketed. FDA’s beef-cattle implant listing describes approved growth implants by animal class and does not list melatonin among the displayed products. A listing should not be treated as a substitute for checking current regulatory status and product labeling.
