What Is Feline Marfan Syndrome? Signs, Diagnosis, and What Researchers Know

A 2026 study described two cat siblings with long limbs, lens luxation, aortic root dilation, and an FBN1 variant. Here is what the finding does—and does not—tell cat owners.

Feline Marfan syndrome is a newly described condition reported in two domestic-cat siblings. The cats had unusually long limbs, lenses displaced in both eyes, and dilation of the aortic root; researchers also identified a potentially disease-causing change in the FBN1 gene. The 2026 report is an important first characterization, but it does not show how common the condition is or establish a routine test or care protocol for cats.

What signs did the reported cats have?

The two littermates, Gary and Shaggy, were noted to have long limbs as kittens. Later veterinary examinations found bilateral lens luxation (displacement of the lenses in both eyes) and aortic root dilation. The researchers describe the eye and aortic findings as cardinal features of Marfan syndrome in these cats.

These are findings in two reported cats, not a validated checklist for diagnosing other cats. Similar signs warrant veterinary evaluation, but they do not by themselves establish Marfan syndrome.

What did the 2026 study find?

Whole-genome sequencing identified the same homozygous splice-region variant in FBN1 in both affected cats: XM_023255387.2:c.2678-3C>A, located three bases upstream of exon 22. Each cat had two copies, one inherited from each parent. FBN1 encodes fibrillin-1, a protein important to connective tissue.

Follow-up cDNA sequencing showed exon 22 skipping in 73% of transcripts in the affected cat studied. The authors describe the variant as hypomorphic: it disrupted RNA splicing but did not eliminate all normal transcript. They interpret the remaining normal transcript as preserving some fibrillin-1 function.

Histopathology of an affected cat’s ascending aorta showed disrupted and fractured elastic-fiber tendrils, consistent with impaired fibrillin-1 function. Together, the clinical observations, genetic result, RNA analysis, and tissue findings support the authors’ characterization of Marfan syndrome in these littermates.

How was feline Marfan syndrome diagnosed in the report?

The report combined clinical assessment with laboratory methods. The investigators examined the cats, used whole-genome sequencing to identify the variant, sequenced cDNA to assess its effect on RNA splicing, and examined aortic tissue histologically.

This describes the methods used in a research report, not a standard clinical workup. The published sources do not establish a routine diagnostic pathway or monitoring schedule for cats with suspected Marfan syndrome. If a cat has concerning eye, limb, or cardiovascular findings, a veterinarian can assess the individual case and decide whether referral to relevant specialists is appropriate.

Is there a genetic test for cats?

The 2026 study identified a specific FBN1 variant in two related cats, but the available sources do not establish that a routine commercial feline test for this variant is available. Research sequencing and specialist interpretation in a published study are not the same as a validated, generally available clinical test.

Cornell’s account of the study quotes senior author Dr. Jacquelyn Evans saying, “The findings provide a foundation for improved veterinary diagnostics.” The article adds that the work “may help develop genetic tests.” That describes potential future value, not an existing test.

How does this compare with earlier feline research?

A 2011 report described primary lens instability in 10 related domestic shorthair cats across three generations. Its pedigree analysis suggested possible dominant inheritance and proposed FBN1 as a candidate, but it did not report a confirmed causal variant. The 2026 report adds a molecularly characterized variant in cats with a broader Marfan-like phenotype, including aortic root dilation.

Study What was reported What it establishes
Payen et al., 2011 Lens instability in 10 related cats over three generations Pedigree evidence suggesting possible dominant inheritance and FBN1 as a candidate; not a confirmed causal variant.
Cook et al., 2026 Two littermates with long limbs, bilateral lens luxation, and aortic root dilation, plus a homozygous FBN1 splice-region variant A molecular and phenotypic characterization of these cases; not a prevalence survey or validation of a routine screening test.

The earlier report should not be read as proof that all feline lens instability is Marfan syndrome, nor as evidence that the 2011 cats carried the variant identified in 2026.

What is not yet known?

  • How often feline Marfan syndrome occurs: the published cases do not establish prevalence in cats.
  • Whether the identified variant explains similar signs in unrelated cats: the 2026 report concerns two littermates.
  • Whether a routine clinical genetic test, standard treatment, or evidence-based monitoring schedule is available: the cited sources do not establish these.

The findings and limitations are described in Cook et al., “A homozygous hypomorphic FBN1 splice region variant in domestic cats with Marfan syndrome,” Scientific Reports, published September 19, 2026, and Cornell University College of Veterinary Medicine’s September 22, 2026 account. The earlier lens-instability report is Payen et al., published July 28, 2011.
