Retracted: The claim that CRISPR-Cas9 produced red fluorescent silk in spiders

A 2025 paper claimed that CRISPR-Cas9-edited spiders produced red fluorescent silk, but the paper was retracted. The publisher says additional tests could not confirm that the reported knockout or knock-in occurred at the intended genomic locations, so the result is not established.

What the retraction means

The retraction is not a minor correction: the authors could not verify the intended edits at their claimed genomic locations. The publisher’s notice says the results could not be verified; it does not give a fuller technical explanation, identify a specific experimental failure, or assign blame.

A visible signal or assay result alone does not establish that a planned DNA sequence was inserted at the intended site. The reported targeted edits were not confirmed in additional tests.

What the withdrawn paper claimed

The retracted paper concerned the cobweb spider Parasteatoda tepidariorum. It reported CRISPR-Cas9 knockout and knock-in work, including insertion of a monomeric red fluorescent protein sequence in a major ampullate silk gene and red-fluorescent silk. These are claims from the retracted paper, not verified findings.

The original article was first published online on April 13, 2025, and is marked retracted by Wiley. The publisher’s retraction notice was first published online on August 21, 2025. It states that additional tests could not confirm that either the knockout or knock-in occurred at the intended genomic location.

What this does—and does not—show

The retraction means this particular paper did not establish that CRISPR produced red fluorescent spider silk. It does not prove that CRISPR editing in spiders is impossible, nor does it settle broader research into spider-silk biomaterials.

Records in PubMed and the University of Bayreuth repository also identify the work as retracted. Its continued appearance in databases does not reverse that status.

How to assess similar science headlines

  1. Check the publisher’s current record for corrections or a retraction.
  2. Separate a paper’s original claim from later verification and editorial notices.
  3. Look for evidence that a genetic edit was confirmed at the intended genomic location, rather than relying only on a visible trait.
  4. Do not treat a proposed application as a demonstrated product or benefit.
